Polycarbonatetrack-etch(PCTE)membranesaretheprimarytypeoffiltersusedinourchambersandinstrumentsystems.Thisisaverythinmembranerangingbetween6-10µminnominalthickness.Thismembranetypeoffersexcellentuniformityinporesizeanddensity,aswellasotherbenefitssuchaslowextractables,lowproteinbinding,andnegligIBLeabsorption.Thesecharacteristicsmakeitideallysuitedforuseinchemotaxisassaysaswellasotherhighpurityscreeningassays.
PVPTreatment–PCTEmembranesareavailablepre-treatedwiththewettingagentpolyvinylpyrrolidone(PVP)whichmakesthesurfacehydrophilic.PVP-treatedmembranesareusedwhenthemediasolutionscannotwetoutthemembranebythemselves.Thisoccursprimarilywhenworkingwithverysmallporesizesorhighviscositymediasolutions.CertaincelltypessuchasneutrophilsdonotadherewelltoPVPtreatedmembranesandtendtodropoffaftermigrating.Whilethisisproblematicwhenquantifyingfiltersonthemembrane,someresearchersusingtheChemoTx®sytemortheMB-series96-wellchambersandquantifyingintheplatefindthistobeanadvantage.Extra-cellularmatrixproteinsdonotadherewelltoPVPtreatedmembraneseither.Ifyourassayrequiressuchcoating,werecommendusingPVP-freemembranes.
FiltersfortheAP48,AC48,AA12andAA10arealsoavailableincellulosenitrate.Thesearemuchthicker(~150µm)fibrousfilters,sometimesreferredtoas“torturouspath”filters.Ratherthancountingcellsthathavemigratedthroughthemembrane,asiscommonlydonewithPCTEmembrane,thesefiltersareclarifiedwithsolventsafterapredeterminedincubation,andahigh-resolutionlightmicroscopeisusedtoobservehowfarintothefilterthecellshavemigrated.Thisisalsoknownasa“leADIngedge”study.Thesefiltershavearecommendedshelflifeofoneyear.Theyarecuttoorder,sopleaseallow2-3daysforprocessing.
