IntheNeuroProbeAA12chemotaxischamberchemoattractantsaddedtothelowerwellsactoncellsinasUSPensionaddedtotheupperwells.Afilterseparatestheupperandlowerwells.Afterincubationthemigratedadherentcellsonthefilterarestainedandcounted.IncomparisonwiththemorecommonlyusedAP48chamber,theAA12haslargervolumeupperandlowerwellsandalargerexposedfilterarea.
TheAA12chamberconsistsofprecision-machinedacrylictopandbottomplates,assemblyhardware,andasiliconegasket.Theplateshave12wellsintwooffsetrowsofsixwellseach.Eachwellinthebottomplateis4.75mmindiameter,holds150µL,andhasaroundedbase.Correspondingholesinthetopplateformtheupperwellswhenthechamberisassembled.Thesetopwellshaveavolumeof100µL.Theexposedfilterareais18mm2.
Asingle25x80mmpieceoffiltermembraneisplacedbetweenthetopandbottomplates,andagasketispositionedoverthefiltertocreateaseal.Narrowraisedrimsaroundmatingsurfacesoftopandbottomwellsenhancetheseal.
Beforethemigratedcellscanbestainedandcounted,thenonmigratedcellsmustbewipedoffthetopsideofthefilter.Anaccessorypack (Stock#P48AP) includesinstrumentsforthispurpose.
TheAA12isdesignedforusewithpolycarbonatefilters,althoughcellulosenitratefiltersmayalsobeused.
Eachchamberisaccompaniedbya protocol foritsuseandcare.FiltersandaccessoriesfortheAA12arethesameasusedwiththeAP48.
